Overview

FOLR1-TCB (also known as RG6150 or RO7209396) is a bispecific T-cell engager developed by Roche. It is designed to simultaneously target Folate Receptor Alpha (FOLR1), which is highly expressed in various solid tumors such as ovarian, lung, and breast cancers, and the CD3 epsilon subunit of the T-cell receptor complex. By binding both targets, FOLR1-TCB facilitates the recruitment and activation of cytotoxic T cells directly to FOLR1-expressing tumor cells, leading to their directed lysis. The molecule utilizes Roche's proprietary T-cell bispecific (TCB) platform, which typically features a 2:1 molecular format (two binding arms for the tumor antigen and one for CD3) to enhance tumor selectivity and potency while minimizing off-target effects.
