FOLR1 TRAILBody is a first-in-class bispecific antibody designed to treat FOLR1-positive solid tumors, specifically ovarian cancer. Developed using the TRAILBody™ platform, it co-targets Folate Receptor Alpha (FOLR1) and Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand Receptor 2 (TRAIL-R2/DR5). The therapeutic mechanism utilizes FOLR1 as a tumor-enriched "clustering anchor" to facilitate the high-order oligomerization of TRAIL-R2 on the tumor cell surface. This clustering triggers rapid caspase-3 activation and induces apoptosis through the extrinsic pathway, bypassing the need for FcγR-mediated clustering which often limits the efficacy of monospecific death receptor agonists. Preclinical data indicates that FOLR1 TRAILBody demonstrates superior anti-tumor activity compared to the antibody-drug conjugate mirvetuximab soravtansine (Elahere) and exhibits a reduced risk of hepatotoxicity compared to traditional TRAIL-R2 agonists.
