Overview
Fontolizumab is a humanized IgG1 kappa monoclonal antibody that targets and neutralizes human interferon gamma (IFN-γ), a proinflammatory cytokine implicated in the pathogenesis of several autoimmune diseases. By binding to IFN-γ and inhibiting its activity and downstream gene expression, fontolizumab acts as an immunosuppressive agent. It was developed primarily for the treatment of Crohn's disease and other autoimmune disorders such as rheumatoid arthritis and psoriasis. Clinical trials demonstrated biological activity (notably reduction in C-reactive protein) but did not show sufficient clinical efficacy to support further development; all programs were discontinued after phase 2 trials[1][2][3][4][5][7].
