Overview

Forchlorfenuron (FCF) is a synthetic cytokinin-like compound primarily known as an agricultural plant growth regulator that has been repurposed in biomedical research as a small molecule inhibitor of the septin cytoskeleton. It functions by binding to septin proteins, such as Septin 7, and disrupting the assembly and stability of septin filaments, which are essential components of the eukaryotic cytoskeleton involved in cytokinesis, cell signaling, and membrane remodeling. In the context of inflammatory bowel disease (IBD), FCF is being investigated for its potential to ameliorate intestinal fibrogenesis. Research, including studies from the Cleveland Clinic, demonstrates that FCF can attenuate the activation of human intestinal myofibroblasts (HIMF) and reduce the production of extracellular matrix (ECM) proteins like collagen and fibronectin. In vivo studies using chronic colitis models have shown that FCF administration can decrease fibrosis scores without significantly affecting inflammation, suggesting a specific anti-fibrotic mechanism of action.
