Overview

Forimtamig is a novel T-cell bispecific antibody designed to treat relapsed or refractory multiple myeloma. It targets G protein-coupled receptor class C group 5 member D (GPRC5D) on malignant plasma cells and CD3 antigen on T cells. By binding simultaneously to these two proteins, forimtamig brings T cells into close proximity with myeloma cells, activating the immune system to destroy the cancerous plasma cells. The drug features a 2+1 format—two high-affinity GPRC5D binding sites and one CD3 binder—and incorporates a silent Fc region (P329G LALA mutation) that reduces toxicity and extends half-life by preventing unwanted innate immune activation while maintaining IgG-like pharmacokinetics. Forimtamig is administered intravenously or subcutaneously in clinical trials and is being evaluated both as monotherapy and in combination with other agents for patients who have failed prior therapies.
