Formalin-inactivated whole-cell non-typeable Haemophilus influenzae (NTHi) is an oral immunotherapeutic and vaccine developed by The University of Newcastle Research Associates (TUNRA) Ltd and Hunter Immunology (later acquired by Bioxyne and licensed to Mariposa Health). The active ingredient consists of lyophilized, formalin-killed whole-cell NTHi (specifically strain NTHi-164), formulated as an enteric-coated tablet (referred to as HI-164OV) to protect against gastric acid. Administered orally, the vaccine targets the mucosal immune system by stimulating gut-associated lymphoid tissue (Peyer's patches). This activates Th17 cells, NTHi-specific T cells, and immunoglobulins (IgG and IgA) that traffic to the respiratory tract, enhancing bacterial clearance and reducing airway inflammation. It is primarily developed for the prevention and treatment of recurrent acute bronchitis and acute exacerbations of chronic obstructive pulmonary disease (COPD) or chronic airway disease.
