Overview

Formononetin is a naturally occurring isoflavone, primarily found in red clover and the traditional Chinese medicinal herb Astragalus membranaceus. It belongs to the class of 7-hydroxyisoflavones, characterized by a methoxy group at position 4'. Formononetin exhibits a range of biological activities, including antioxidant, anti-inflammatory, anti-cancer, anti-apoptotic, antiviral, antibacterial, and estrogenic effects. Mechanistically, it modulates multiple cellular signaling pathways such as PI3K/AKT, MAPK/ERK/JNK (mitogen-activated protein kinase/extracellular signal-regulated kinase/c-Jun N-terminal kinase), STAT (signal transducer and activator of transcription), and nuclear factor kappa B (NF-κB). In cancer models it induces apoptosis via mitochondrial pathways involving Bax/Bcl-2/caspase-3 proteins and causes cell cycle arrest by regulating cyclins. In neurodegenerative disease models like Alzheimer’s disease it may inhibit amyloid-beta/RAGE interaction and reduce cytokine levels through modulation of Nrf2 signaling. Formononetin has been investigated in clinical trials for postmenopausal osteopenia[1][5][6][8].
