Drug intelligence / Profile preview
formyl peptide receptor 1 siRNA is a small interfering RNA (siRNA) therapeutic designed to silence the expression of formyl peptide receptor 1 (FPR1). In preclinical models of sepsis-associated encephalopathy (SAE), FPR1 was identified as a key driver of pro-inflammatory microglial polarization and neuroinflammation. To overcome blood-brain barrier (BBB) delivery challenges, researchers developed an intranasal delivery system utilizing piezoelectric short fibers loaded with FPR1-siRNA@MG1-LNPs. This system converts mechanical energy from respiratory airflow and nasal ciliary beating into a localized microelectric field, enabling the therapeutic genes to bypass the BBB via electro-responsive active migration. Silencing of FPR1 suppresses microglial pyroptosis, reduces the release of inflammatory mediators, mitigates neuronal injury, and ameliorates cognitive dysfunction.
