Overview

formylmethionyl-leucyl-phenylalanine is a synthetic tripeptide and the prototypical representative of formylated oligopeptides, often referred to as fMLF or fMLP. It acts as a potent chemotactic factor for polymorphonuclear leukocytes (PMN) and is a well-characterized agonist at the formyl peptide receptor family (FPR1, FPR2, and FPR3), which are G protein-coupled receptors on leukocytes. fMLF is released by bacteria and serves as a signal for leukocyte migration to infection or inflammation sites, activating both chemotaxis and degranulation. Mechanistically, fMLF interacts with FPR1 as a high-affinity agonist, leading to activation of multiple leukocyte responses including superoxide generation, lysosomal enzyme release, and macrophage activation. It has been extensively used as a tool compound in immunology and inflammation research, but is not an approved therapeutic drug[1][2][3][5][8].
