Forsythia velutina Nakai extract (FVE) is a natural product derived from the plant Forsythia velutina Nakai. It has demonstrated promising therapeutic potential in the treatment of atopic dermatitis (AD), a chronic inflammatory skin disease. Mechanistically, FVE exerts anti-inflammatory effects by inhibiting multiple cell types involved in AD pathogenesis, including basophils, macrophages, keratinocytes, and T cells. The extract downregulates inflammatory mediators and reactive oxygen species production in macrophages via modulation of TLR4 and NRF2 pathways; reduces basophil degranulation and type 2/pro-inflammatory cytokine expression by perturbing FcεRI signaling; inhibits type 2 cytokine expression in activated T cells; upregulates tight junction molecules in damaged keratinocytes; and downregulates leukocyte attractants such as IL-33. In animal models of AD, FVE improved skin integrity and reduced inflammation more effectively than dexamethasone. Dimatairesinol, a lignan dimer found within the extract, was identified as one of its most potent anti-inflammatory constituents[1][3][4].
