Overview

Forsythoside I is a phenylethanoid glycoside, likely derived from the medicinal plant *Forsythia suspensa*, that acts as a small molecule agonist of the small GTPase **RAC1**. Research presented at the AHA 2025 meeting identifies it as a potential therapeutic for **aortic aneurysm and dissection (AAD)**. The compound specifically binds to the Q61 site of RAC1, promoting its activity and counteracting the inhibitory effects of elevated MST1 expression. By restoring RAC1 activation and subsequent FLNB-mediated vascular smooth muscle cell contractility, Forsythoside I has demonstrated significant therapeutic efficacy and a high safety profile in preclinical animal models of AAD.
