FORT-101 is a next-generation antibody-drug conjugate (ADC), specifically a Degrader-Antibody Conjugate (DAC), being developed by Fortitude Biomedicines for the treatment of cancer. It utilizes the company's proprietary GLUE-DAC™ platform, which employs molecular glue payloads to induce targeted protein degradation. FORT-101 is designed to promote the degradation of key transcriptional coactivators that are essential for the survival and proliferation of cancer cells. By combining the precision of antibody-mediated delivery with the catalytic power of protein degradation, FORT-101 aims to overcome resistance mechanisms associated with traditional cytotoxic ADCs and expand the therapeutic window. As of early 2026, the program is in the discovery stage.
