Fosamprenavir + sodium alginate is a combination drug that pairs fosamprenavir (an FDA-approved HIV protease inhibitor) with sodium alginate (a natural polymer with muco-adhesive properties). This combination is being developed as a treatment for laryngopharyngeal reflux (LPR) and potentially other reflux conditions. The therapeutic approach is based on fosamprenavir's ability to bind to and inhibit pepsin, which is a primary cause of damage in LPR. Pepsin is detected in the airways of LPR patients and causes laryngeal inflammation independent of gastric acid. Sodium alginate is added to the formulation to increase muco-adhesion and potentially provide sustained release of the active ingredient. This combination represents a drug repurposing strategy, as fosamprenavir is already FDA-approved for HIV treatment, which allows for a faster path to clinical trials for this new indication. The combination is being developed in both oral suspension and potentially inhaled formulations.
