**Fosigotifator sodium tromethamine** is the oral monosodium phosphate salt and tromethamine presentation of fosigotifator, an investigational, brain-penetrant small-molecule activator of eukaryotic translation initiation factor 2B. It stabilizes and increases the activity of eIF2B, including eIF2B complexes carrying pathogenic vanishing white matter disease variants, thereby attenuating maladaptive chronic integrated stress response signaling and supporting restoration of protein synthesis.
