**Fosigotifator sodium tromethamine** is the oral monosodium phosphate salt and tromethamine presentation of fosigotifator, an investigational, brain-penetrant small-molecule activator of eukaryotic translation initiation factor 2B. It stabilizes and increases the activity of eIF2B, including eIF2B complexes carrying pathogenic vanishing white matter disease variants, thereby attenuating maladaptive chronic integrated stress response signaling and supporting restoration of protein synthesis. Calico developed fosigotifator in collaboration with AbbVie for vanishing white matter disease; it is also being evaluated or has been evaluated in amyotrophic lateral sclerosis, major depressive disorder, and Cree leukoencephalopathy.
