Overview

Foslip is a third-generation photosensitizer consisting of a liposomal formulation of temoporfin (meta-tetra(hydroxyphenyl)chlorin or mTHPC). Developed by Biolitec, it utilizes unilamellar dipalmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidylglycerol (DPPG) liposomes to encapsulate the active small molecule. This formulation is designed to improve the pharmacokinetics and tumor selectivity of temoporfin while minimizing common side effects associated with the standard formulation (Foscan), such as prolonged skin photosensitivity. When irradiated with red light at 652 nm, Foslip generates reactive oxygen species (ROS) and singlet oxygen, which cause targeted destruction of tumor cells and surrounding vasculature. It has been evaluated in preclinical and Phase I clinical trials for the treatment of various cancers, including head and neck and colorectal malignancies, as well as for applications in wound healing.
