Overview

Fourth-Generation CAR-T is an investigational autologous chimeric antigen receptor T-cell therapy developed by The Third Affiliated Hospital of Southern Medical University. This fourth-generation (4G) CAR-T platform, also known as TRUCKs (T cells redirected for universal cytokine killing), is designed to treat various hematologic malignancies by targeting specific antigens such as B-cell maturation antigen (BCMA) for multiple myeloma, CD19 for B-cell lymphoma, or CD7 for other T-cell malignancies. These 4G CAR-T cells are engineered with additional genetic elements, such as cytokine-expressing sequences, to enhance their expansion, persistence, and ability to overcome the immunosuppressive tumor microenvironment compared to earlier generations.
