Fovista + bevacizumab is an investigational combination therapy for neovascular (wet) age-related macular degeneration (AMD). Fovista (pegpleranib, also known as E10030) is a pegylated aptamer that targets and inhibits platelet-derived growth factor-BB (PDGF-BB), disrupting pericyte support of abnormal blood vessels. Bevacizumab is a monoclonal antibody that binds to and neutralizes vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis. The rationale for combining these agents is to simultaneously block two key pathways involved in the formation and maintenance of abnormal choroidal neovascularization in wet AMD—VEGF-driven endothelial cell proliferation/leakage and PDGF-mediated pericyte recruitment/stabilization. Clinical trials have evaluated this combination to determine if dual inhibition offers superior visual outcomes compared to anti-VEGF monotherapy[2][3][4][5][6].
