Overview

Fowlpox-B7.1 (rF-B7.1) is a recombinant, non-oncolytic fowlpox virus-based cancer vaccine developed by the National Cancer Institute (NCI). The vaccine consists of a fowlpox virus vector engineered to express the human T-cell costimulatory molecule B7.1 (CD80). Its primary mechanism of action involves the delivery of B7.1 directly into the tumor microenvironment via intratumoral injection, which provides the necessary costimulatory signal to enhance T-cell activation and promote a cell-mediated immune response against tumor cells. Because the fowlpox virus is an avian virus that does not replicate in mammalian cells, it provides a high safety profile with minimal risk of systemic infection. Fowlpox-B7.1 has been evaluated in Phase I clinical trials for the treatment of metastatic solid tumors, including melanoma and colon adenocarcinoma, often serving as a comparator or component in studies involving other costimulatory molecules like those in the TRICOM (B7.1, ICAM-1, and LFA-3) platform.
