Overview

fowlpox virus-gp100 is a recombinant viral vaccine vector designed for the immunotherapy of malignant melanoma. It utilizes a live, attenuated, non-replicating fowlpox virus (a member of the Avipoxvirus genus) engineered to express the human gp100 (Pmel17) gene, a well-characterized melanoma-associated antigen. The primary therapeutic strategy involves a heterologous prime-boost regimen, where a recombinant vaccinia virus-gp100 (rV-gp100) is typically used for priming and the fowlpox-based vector is used for boosting. This approach is intended to bypass anti-vector immunity and maximize the expansion of gp100-specific CD8+ cytotoxic T lymphocytes, which then target and destroy melanoma cells. Clinical development has primarily focused on advanced or metastatic melanoma, often in combination with cytokines like interleukin-2 (IL-2) or costimulatory molecules.
