Overview

The FOXM1-targeted peptide vaccine is a therapeutic cancer vaccine currently under investigation for the treatment of triple-negative breast cancer (TNBC). The vaccine targets the transcription factor Forkhead Box M1 (FOXM1), which is a known driver of immune evasion and suppression in the tumor microenvironment. FOXM1 acts by downregulating MHC-I expression and the STING pathway, thereby disrupting NK and CD8+ T cell-mediated cytotoxicity. By utilizing immunogenic peptides identified from FOXM1-deficient cells, the vaccine aims to stimulate robust T cell responses, remodel the immune landscape to increase memory T cell populations, and provide durable antitumor protection. Research presented at ASCO 2026 highlights its potential to improve long-term disease control in TNBC patients, particularly when combined with immune checkpoint blockade.
