FOXO1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the Forkhead box protein O1 (FOXO1) transcription factor through the RNA interference (RNAi) pathway. FOXO1 is a key regulator of various cellular processes, including apoptosis, cell cycle progression, and metabolism. In oncological research, FOXO1 siRNA is utilized to investigate the transcription factor's role in promoting cancer cell survival and progression. For example, studies in prostate and melanoma cell lines have shown that FOXO1 silencing leads to a significant reduction in the expression of Protein Kinase C-ι (PKC-ι), an oncogene that helps cancer cells evade senescence and apoptosis. This suggests that FOXO1 acts as an upstream activator of PKC-ι, potentially via the NF-κB pathway, highlighting its potential as a therapeutic target in malignancies.
