FOXP3 AUMsilence ASO is a next-generation antisense oligonucleotide (ASO) developed by AUM Biotech (AUM LifeTech) designed to target FOXP3 mRNA. The compound utilizes a 2'-deoxy-2'-fluoro-beta-D-arabinose (FANA) sugar modification and a phosphorothioate backbone, structured as a DNA segment flanked by FANA segments (a gapmer configuration). By employing RNA silencing to downregulate the expression of the FOXP3 transcription factor, the drug aims to alleviate the immunosuppressive activity of regulatory T cells (Tregs) within the tumor microenvironment. Preclinical studies have demonstrated significant reduction of FOXP3 mRNA levels in peripheral blood mononuclear cells (PBMC) and clinical samples from patients with lung cancer, mesothelioma, and melanoma, supporting its development as a potential cancer immunotherapy.
