Foxy-5 is a formylated hexapeptide that mimics the activity of WNT5A, a non-canonical Wnt ligand. It is designed as an anti-metastatic agent for cancers characterized by low or absent WNT5A expression. Mechanistically, Foxy-5 binds to and activates Frizzled receptors (specifically Frizzled-2 and Frizzled-5), stimulating WNT/Calcium signaling pathways without activating β-catenin-dependent signaling. This activation impairs tumor cell migration and invasion but does not affect proliferation or apoptosis. The drug has been investigated in clinical trials for metastatic cancers including prostate cancer, colorectal cancer (notably in patients with low WNT5A expression), metastatic colon cancer, metastatic breast cancer, and metastatic prostate cancer[1][2][3][6][7]. Developed by the company WntResearch[7], Foxy-5 is administered intravenously and has reached phase II clinical development for colorectal cancer[7].
