FP-385 is a gut-restricted, small molecule inhibitor of the LolCDE lipoprotein transport system, currently under development by Flightpath Biosciences. It is designed as a precision antimicrobial that selectively targets Gram-negative pathogens such as Adherent-Invasive *Escherichia coli* (AIEC), *Klebsiella pneumoniae*, and *Enterobacter cloacae*, which are linked to the pathogenesis of inflammatory bowel disease (IBD) and Crohn's disease. The LolCDE complex is an essential ATP-binding cassette (ABC) transporter that facilitates the trafficking of lipoproteins from the inner membrane to the outer membrane; its inhibition leads to bacterial cell wall disruption and death. By remaining localized in the gastrointestinal tract, FP-385 aims to eliminate disease-driving bacteria while minimizing systemic exposure and preserving the commensal microbiome.
