Drug intelligence / Profile preview
FP-rTTHC (also known as FP8v1-rTTHC or VRC-HIVVCP0108-00-VP) is an investigational HIV-1 vaccine candidate developed by the Vaccine Research Center (VRC) of the National Institute of Allergy and Infectious Diseases (NIAID). It consists of the N-terminal eight residues of the HIV-1 envelope glycoprotein gp41 fusion peptide (FP8) conjugated to a recombinant tetanus toxoid heavy chain fragment C (rTTHC) carrier protein via a chemical linker (such as sulfo-SIAB). FP-rTTHC is designed to act as a priming immunogen that targets a highly conserved site of vulnerability on the HIV-1 envelope, initiating the development of fusion peptide-directed broadly neutralizing antibodies (bnAbs). It is typically evaluated in prime-boost regimens followed by boosts with prefusion-stabilized Env trimers (such as Trimer 4571 and Trimer 6931). Clinical development of FP-rTTHC was evaluated in the Phase 1 HVTN 303 trial (NCT05470400), which was terminated due to safety concerns related to the Adjuplex adjuvant.
