FP253 + fludarabine is a gene-directed enzyme prodrug therapy (GDEPT) combination developed by Biotech Equity Partners for the treatment of prostate cancer. The therapy involves a two-step process: first, FP253, an ovine atadenovirus vector carrying the *Escherichia coli* purine nucleoside phosphorylase (PNP) gene under a prostate-specific promoter, is injected directly into the prostate. Second, the prodrug fludarabine phosphate is administered systemically. The PNP enzyme expressed in the prostate cells converts the relatively non-toxic fludarabine into the highly cytotoxic metabolite 2-fluoroadenine. This localized conversion is designed to concentrate the chemotherapeutic effect within the prostate tumor, thereby maximizing anti-tumor activity while minimizing systemic toxicity and damage to adjacent healthy tissues.
