Overview

FPA-306 is a novel small molecule inhibitor of cyclooxygenase-2 (COX-2) specifically investigated for its anti-tumor activity in pancreatic cancer. It functions by binding to the COX-2 active site, thereby inhibiting the synthesis of prostaglandin E2 (PGE2), a key mediator in cancer cell proliferation and survival. In preclinical models, FPA-306 has demonstrated the ability to inhibit the growth of COX-2-overexpressing pancreatic cancer cells (e.g., BxPC-3) and induce apoptosis. This effect is associated with the downregulation of NF-kappaB signaling and its downstream targets, such as Bcl-2 and survivin. FPA-306 appears selective for COX-2-expressing cells, showing no significant effect on COX-2-negative cell lines such as MIAPaCa.
