Overview

FPA157 is an investigational, Fc‑engineered monoclonal antibody targeting the chemokine receptor CCR8, originally developed by Five Prime Therapeutics and now under Amgen’s oncology portfolio following acquisition. It is designed with enhanced antibody-dependent cell-mediated cytotoxicity (ADCC/eADCC) to selectively deplete CCR8-positive tumor-infiltrating regulatory T cells (Tregs) while sparing peripheral Tregs, thereby relieving local immunosuppression and amplifying anti-tumor immune responses in solid tumors.[3][4][5][6][10][7] Preclinical models have shown potent natural killer (NK) cell–dependent killing of CCR8+ Tregs and anti-tumor activity, and the molecule has progressed from a preclinical immunomodulator program toward first-in-human development for advanced solid tumors, including in combination with PD‑1 blockade.[1][3][4][5][6][7]
