FPE021

FPE021 is a trispecific T-cell engager (TCE) developed by Guangdong Fapon Biopharma for the treatment of solid tumors. The therapeutic is designed to simultaneously bind CD3 and CD28 on T-cells and Cadherin-17 (CDH17) on tumor cells. By integrating the CD28-mediated 'second signal' (costimulation) alongside the CD3-mediated 'first signal' (activation), FPE021 aims to enhance T-cell proliferation, mitigate apoptosis, and prevent T-cell exhaustion, addressing key barriers to TCE efficacy in solid tumor microenvironments. Preclinical studies presented at AACR 2026 demonstrated that FPE021 provides potent tumor suppression in vivo and superior cytotoxicity compared to traditional bispecific TCEs, while maintaining a favorable safety profile by avoiding non-specific T-cell activation and fratricide.
