FPE026 is a first-in-class, next-generation tri-specific T cell engager (TCE) designed to target Cadherin-17 (CDH17) on tumor cells while simultaneously binding CD3 and NKG2D on T cells. Developed by FineImmune, it is engineered for CD8-selective activation. Unlike traditional TCEs that activate all T cell subsets, FPE026 specifically targets CD8+ T cells because NKG2D is highly expressed on CD8+ T cells but rarely on CD4+ T cells or regulatory T cells (Tregs). This selectivity aims to reduce toxicities like cytokine release syndrome (CRS) by inducing lower levels of pro-inflammatory cytokines (TNF-α, IL-6) and prevents the inadvertent activation of immunosuppressive Tregs. The inclusion of the NKG2D binding domain provides a costimulatory signal that restores CD8+ effector function even in the absence of CD4+ T cell help.
