Overview

FPI-1848 is an investigational targeted alpha therapy (TAT) radiopharmaceutical developed by Fusion Pharmaceuticals (now a subsidiary of AstraZeneca). It consists of a targeting moiety specific for the Insulin-like Growth Factor 1 Receptor (IGF-1R) conjugated to the alpha-emitting radioisotope Actinium-225 (^225Ac) via Fusion's proprietary "Fast-Clear" linker technology. IGF-1R is frequently overexpressed in various solid tumors and plays a key role in tumor growth and survival. By targeting this receptor, FPI-1848 delivers high-energy alpha particles directly to the tumor microenvironment, causing lethal double-stranded DNA breaks in malignant cells while the Fast-Clear linker facilitates rapid systemic elimination of the non-bound drug to reduce radiation exposure to healthy tissues. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
