FPV-Brachyury is a recombinant cancer vaccine that uses a fowlpox virus vector engineered to express the human brachyury gene. It is administered as part of a prime-boost immunotherapy regimen, typically following priming with MVA-BN-Brachyury (a modified vaccinia Ankara vector expressing brachyury and costimulatory molecules). The primary mechanism is to stimulate the immune system—specifically T-cell responses—against tumor cells overexpressing the transcription factor brachyury, which is implicated in tumor progression, metastasis, and resistance to therapy. This approach aims to enhance anti-tumor immunity in patients with advanced solid tumors such as chordoma and metastatic breast cancer.
