Overview

FR054 is a small molecule inhibitor of phosphoglucomutase 3 (PGM3), a key enzyme in the hexosamine biosynthetic pathway (HBP). The HBP is responsible for the synthesis of UDP-GlcNAc, a critical substrate for protein O-GlcNAcylation and the production of extracellular matrix (ECM) macromolecules. By inhibiting PGM3, FR054 reduces HBP flux, which has been shown to attenuate fibroblast-to-myofibroblast differentiation and decrease the expression of fibrotic markers such as fibronectin and collagen. This mechanism is currently being investigated as a potential anti-fibrotic strategy for Idiopathic Pulmonary Fibrosis (IPF). Additionally, FR054 has been studied in oncology for its ability to disrupt glycosylation-dependent pro-survival signaling and induce apoptosis in various cancer cell lines.
