FraAP (Fra1 antagonist peptide) is a preclinical peptide therapeutic developed by Sapience Therapeutics using its proprietary SPEARs™ (Stabilized Peptides Engineered Against Regulation) platform. The drug is designed to target traditionally 'undruggable' transcription factors by disrupting specific intracellular protein-protein interactions. Specifically, FraAP targets Fra1 (FOSL1) and disrupts its interaction with cJun within the Activator Protein 1 (AP-1) transcription factor complex. By preventing the formation of the cJun/Fra1 AP-1 complex, FraAP inhibits oncogenic transcriptional programs that drive cancer cell proliferation, invasion, and survival. Preclinical studies have demonstrated its efficacy in inducing tumor regression in head and neck squamous cell carcinoma (HNSCC) models and reducing cancer cell invasion across various solid tumor types.
