Freeze-dried black raspberries (BRBs) are a botanical product derived from the fruit of *Rubus occidentalis*, which has been extensively investigated for its chemopreventive and anti-inflammatory properties, particularly by researchers at Ohio State University. BRBs contain a complex profile of bioactive compounds, including anthocyanins (such as cyanidin-3-rutinoside), ellagic acid, quercetin, vitamins, and minerals. These components work synergistically to modulate multiple signaling pathways involved in carcinogenesis and chronic inflammation. Key mechanisms include the suppression of the PI3K/AKT/mTOR, NFκB, and MAPK pathways, as well as the inhibition of pro-inflammatory and pro-angiogenic mediators like COX-2, iNOS, and VEGF. Additionally, BRBs have been shown to influence epigenetic regulation by reversing aberrant DNA methylation in the Wnt signaling pathway through the downregulation of enzymes such as DNMT3B and HDACs. Clinical development has explored BRBs in various formulations, including oral powders, lozenges (troches), and topical gels, for indications such as esophageal cancer, oral intraepithelial neoplasia, and colorectal cancer.
