FRGxCTLA4 is a bispecific monoclonal antibody being developed by Ocean Biomedical and Aesther Healthcare for the treatment of cancer, including glioblastoma and malignant melanoma. The drug simultaneously targets two biological targets: Chitinase-3-like protein 1 (CHI3L1, also known as YKL-40) and Cytotoxic T-lymphocyte-associated protein 4 (CTLA4), an immune checkpoint protein. As a bispecific antibody, FRGxCTLA4 works by inducing T cell differentiation into CD8+ cytotoxic T cells, leading to tumor cell death and preventing tumor metastasis. In preclinical studies using murine models, FRGxCTLA4 demonstrated synergistic effects, blocking melanoma metastasis to the lungs by over 90% and showing impressive tumor cell killing activity in glioblastoma. The drug represents a novel approach to cancer immunotherapy by combining anti-CHI3L1 targeting with immune checkpoint inhibition.
