FRGxPD-1 is a bispecific antibody designed to simultaneously target Fibroblast Activation Protein (FAP) and Programmed Cell Death Protein 1 (PD-1). Developed by Frontier Biotechnologies using their proprietary FRG (FAP-targeted Receptor-agonist/antagonist Generation) platform, this agent is engineered to localize PD-1 blockade to the tumor microenvironment by binding to FAP, which is highly expressed on cancer-associated fibroblasts (CAFs). By anchoring to FAP, FRGxPD-1 facilitates the concentration of the checkpoint inhibitor within the tumor stroma, potentially enhancing the activation of tumor-infiltrating lymphocytes while minimizing systemic immune-related adverse events. This dual-targeting approach aims to overcome the limitations of traditional PD-1 inhibitors in tumors with dense stroma or low baseline immune infiltration.
