FRM-0334 is an orally available, small molecule inhibitor of histone deacetylase (HDAC) enzymes. HDACs are key regulators of gene expression through epigenetic mechanisms; by removing acetyl groups from histones, they repress gene transcription. FRM-0334 was developed to increase the expression of progranulin by inhibiting HDACs, which may be neuroprotective in diseases where progranulin deficiency is implicated. The drug has demonstrated good brain penetration and safety in preclinical and early clinical studies. It was primarily developed for frontotemporal dementia (FTD) associated with mutations in the granulin (GRN) gene, a condition characterized by reduced progranulin levels leading to neurodegeneration[1][2][6].
