Overview

Frovocimab (LY3015014) + statin is a combination therapy used for lipid-lowering in patients with primary hypercholesterolemia, especially those not adequately controlled with statin therapy alone. Frovocimab is a humanized IgG4 monoclonal antibody targeting proprotein convertase subtilisin/kexin type 9 (PCSK9), which increases LDL receptor recycling and reduces plasma LDL-C levels. Statins inhibit HMG-CoA reductase, reducing cholesterol biosynthesis and upregulating LDL receptors. The combination results in additive or synergistic reduction in LDL-C and related atherogenic lipoproteins. Primary clinical studies focused on robust LDL-C lowering in patients receiving maximally tolerated statin therapy, with or without ezetimibe, and demonstrated durable LDL-C reductions. Frovocimab is developed by Eli Lilly and Company and studied in several multinational Phase 1–2 clinical trials.
