Overview

Fruquintinib + PD-1 inhibitor is a combination therapy being evaluated as a first-line maintenance treatment for patients with HER2-negative advanced gastric or gastroesophageal junction (GEJ) adenocarcinoma. Fruquintinib is a highly selective and potent oral small molecule inhibitor of VEGFR-1, -2, and -3, which blocks tumor-induced angiogenesis. The PD-1 inhibitor component (which may include nivolumab, sintilimab, tislelizumab, or pembrolizumab) is a monoclonal antibody that targets the programmed cell death protein 1 receptor on T-cells, preventing its interaction with ligands PD-L1 and PD-L2 to restore anti-tumor immune responses. This regimen is typically administered after patients achieve disease control with initial platinum-based chemotherapy.
