Overview

Fruquintinib + sintilimab + SOX is a combination regimen used as conversion therapy for patients with initially unresectable, locally advanced or metastatic gastric cancer (GC) or gastroesophageal junction cancer (GEJC). Fruquintinib is an oral, highly selective inhibitor of vascular endothelial growth factor receptors 1, 2, and 3 (VEGFR1/2/3), which inhibits tumor angiogenesis. Sintilimab is a monoclonal antibody targeting programmed death-1 (PD-1), functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. The SOX regimen consists of S-1 (an oral fluoropyrimidine derivative) and oxaliplatin (a platinum-based chemotherapeutic agent), both of which are cytotoxic agents commonly used in gastrointestinal cancers. This combination leverages the synergistic effects between antiangiogenic therapy, immunotherapy, and chemotherapy to improve resectability rates and clinical outcomes in advanced gastric cancer[6][8].
