fruquintinib + SOX is a combination neoadjuvant therapy regimen being evaluated for the treatment of locally advanced gastric and gastroesophageal junction (GEJ) adenocarcinoma. The regimen consists of fruquintinib, a highly selective and potent oral tyrosine kinase inhibitor of VEGFR-1, 2, and 3, combined with the SOX chemotherapy regimen (S-1 plus oxaliplatin). Fruquintinib, developed by HUTCHMED, inhibits tumor angiogenesis, while SOX provides cytotoxic effects through DNA cross-linking (oxaliplatin) and inhibition of thymidylate synthase (S-1). This specific combination is being investigated in a Phase II clinical trial sponsored by Guangxi Medical University to improve R0 resection rates and long-term survival in patients with stage III disease.
