FS120 is a first-in-class, dual-agonist tetravalent bispecific antibody that targets CD137 (also known as 4-1BB, TNFRSF9) and OX40 (also known as CD134, TNFRSF4), both of which are members of the tumor necrosis factor receptor superfamily. By simultaneously agonizing these two receptors on activated T cells and NK cells, FS120 aims to enhance anti-tumor immune responses. Unlike many other antibodies targeting these pathways, FS120 is designed to be FcγR-null and activates its targets through bispecific crosslinking rather than Fcγ receptor engagement, reducing toxicity risks associated with traditional monoclonal antibodies. Preclinical studies have shown that FS120 can activate both CD4+ and CD8+ T cells independently of FcγR interaction, promote cytokine production, delay tumor growth in "cold" tumors resistant to immunotherapy, and synergize with PD-1 inhibitors. The drug was originally developed by F-star Therapeutics and is now being advanced by invoX Pharma for the treatment of advanced malignancies including solid tumors[3][5][6][7][8].
