FT-105

FT-105 is a potent, selective, and orally bioavailable small molecule glucocorticoid receptor modulator (SGRM) developed by Forma Therapeutics. It was designed to act as an antagonist of the glucocorticoid receptor (GR), a nuclear receptor that can drive resistance to androgen receptor (AR) targeted therapies in metastatic castration-resistant prostate cancer (mCRPC). By inhibiting GR signaling, FT-105 was intended to restore sensitivity to AR antagonists or provide therapeutic benefit in tumors where GR expression is upregulated as a bypass mechanism. Clinical development included Phase 1 trials for advanced solid tumors and mCRPC, but the program was ultimately terminated for strategic business reasons.
