FT-ONO1 is an investigational drug candidate derived from induced pluripotent stem cell (iPSC) technology and engineered as a T cell immunotherapy. It is being developed jointly by Fate Therapeutics and Ono Pharmaceutical, primarily for the treatment of hematologic malignancies. FT-ONO1 utilizes CAR-targeted, TCR-null CD8αβ+ T cells generated from iPSCs. The approach aims to provide off-the-shelf, allogeneic cell therapy targeting cancer cells, leveraging the advantages of iPSC-derived immune effector cells for scalable and consistent manufacturing. The mechanism of action is based on engineered T cells targeting specific tumor antigens via Chimeric Antigen Receptor (CAR) technology, and the use of TCR-null cells aims to reduce the risk of graft-versus-host disease in recipients[1][15].
