FT516 + rituximab

FT516 + rituximab is an investigational combination therapy for relapsed or refractory B-cell lymphoma, consisting of FT516—an off-the-shelf natural killer (NK) cell cancer immunotherapy derived from a clonal master induced pluripotent stem cell (iPSC) line engineered to express a novel high-affinity, non-cleavable CD16 Fc receptor—and rituximab, a CD20-targeted monoclonal antibody. FT516 is designed to maximize antibody-dependent cellular cytotoxicity (ADCC), enhancing the NK cell-mediated killing of antibody-coated lymphoma cells. Rituximab binds to CD20 on B-cells, enabling the recruitment of immune-effector mechanisms including ADCC. The combination regimen is being studied in patients with relapsed or refractory B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma, particularly in patients who have failed multiple prior therapies, including CAR T-cell therapy. The regimen is designed to be administered in the outpatient setting and includes lymphodepleting chemotherapy with cyclophosphamide and fludarabine followed by a single dose of rituximab and multiple doses of FT516 with IL-2 support[1][2][3][4].
