Overview

FT516 + avelumab is an investigational **combination therapy** comprised of FT516, an engineered “off-the-shelf” natural killer (NK) cell product derived from genetically modified human induced pluripotent stem cells (iPSC), and avelumab, a fully human IgG1 monoclonal antibody targeting programmed death-ligand 1 (PD-L1). FT516 has enhanced expression of high-affinity CD16a and resistance to ADAM17 cleavage to increase NK cell-mediated cytotoxicity and antibody-dependent cell-mediated cytotoxicity (ADCC). Avelumab acts as a PD-L1 checkpoint inhibitor and enables further ADCC through engagement of immune effector cells. The combination is designed to synergistically target and eliminate cancer cells via both immune checkpoint blockade and boosted NK cell activity, particularly in advanced solid tumors[1][5][6].
