Overview

FT522 is an investigational, off-the-shelf, iPSC-derived chimeric antigen receptor (CAR) natural killer (NK) cell therapy developed by Fate Therapeutics. It is engineered to target CD19-expressing cells and incorporates a novel alloimmune defense receptor (ADR) technology designed to enhance the potency of cell therapy and enable treatment without the need for conditioning chemotherapy. The ADR technology allows FT522 to persist and function in patients with fully intact immune systems by targeting Tumor necrosis factor receptor superfamily member 9 (4-1BB)+ immune cells, promoting functional persistence while selectively depleting pathogenic CD19+ B cells. Preclinical data demonstrate rapid and deep B-cell depletion, enhanced persistence, and elimination of alloreactive host immune cells. FT522 is being evaluated in Phase 1 clinical trials for relapsed/refractory B-cell lymphoma as well as for a range of B cell-mediated autoimmune disorders.
