Overview

FT536 + pembrolizumab is an investigational combination therapy comprising FT536, an off-the-shelf, gene-edited, chimeric antigen receptor (CAR) natural killer (NK) cell therapy, and pembrolizumab, a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor monoclonal antibody. FT536 is derived from an induced pluripotent stem cell (iPSC) line and is engineered to target the α3 domain of MICA and MICB, stress ligands upregulated on many tumor cells to enable immune evasion. Additional genetic modifications include a modified CD16 receptor for enhanced engagement with tumor-targeting antibodies, IL-15 receptor fusion for increased NK activity, and CD38 knockout for improved persistence. Pembrolizumab blocks PD-1, restoring T cell function and enhancing antitumor immune response. The combination is being evaluated in Phase 1 studies for advanced solid tumors, leveraging both direct cytotoxicity (FT536) and relief of immune inhibition (pembrolizumab)[1][3][4].
